Plectin is a cytoskeletal protein, which is essential for normal function of the glomerular slit diaphragm. It was shown that plectin binds the tight junction protein occludin. Here we show that in vitro plectin binds several tight junction proteins, including occludin. In addition to occludin, plectin also binds zonula occludens-1 (ZO-1). To determine if plectin can influence the localization of occludin, a C-terminal plectin fragment containing the actin-binding and bundling domain, termed P1, was expressed in Madin-Darby canine kidney (MDCK) cells. P1 colocalized with occludin at the cell periphery. Expression of P1 did not affect the overall distribution of occludin. P1 was able to bind occludin and ZO-1 but not claudin-4. In contrast, the myc-epitope tagged full-length plectin colocalized with occludin and ZO-1. The importance of the actin-binding domain for plectin's influence on the localization of occludin was shown by the reduced colocalization of occludin with plectin in the presence of an actin-binding domain-deficient mutant. The ability of plectin to influence the localization of occludin may play a role in the recruitment of occludin to the tight junction.
